Document ID: EE-MM-2023-WQ-001

Prepared By: Senior Environmental Engineer

Location: Philippines Manila (Metro Manila Region)

Date: October 24, 2023

Subject: Standard Operating Procedure for Sampling and Analysis of Surface Water in the Pasig River Basin

This Experiment Protocol is designed for the use of the Environmental Engineer and technical staff operating within the Philippines Manila metropolitan area. The primary objective is to establish a rigorous methodology for assessing the physicochemical and biological quality of surface water bodies, specifically focusing on the Pasig River and its tributaries.

Given the high density of urbanization and industrial activity in Manila, this protocol aims to quantify pollutant loads, including heavy metals, biological oxygen demand (BOD), and fecal coliforms. The data gathered will support compliance with the Philippine Clean Water Act of 2004 (Republic Act No. 9275) and assist local government units in remediation efforts.

This protocol applies to all field sampling and laboratory analysis conducted by the Environmental Engineer in the Philippines Manila region. It covers:

  • Pre-sampling site assessment.
  • Collection of water samples from designated stations.
  • Preservation and transportation of samples.
  • Laboratory analysis parameters.
  • Data reporting and quality assurance.

Working in the Philippines Manila environment presents specific hazards, including biological contaminants and traffic risks. The Environmental Engineer must ensure the following safety measures are strictly observed:

  • PPE: Nitrile gloves, safety goggles, waterproof boots, and high-visibility vests are mandatory.
  • Health: Due to potential exposure to pathogens, all personnel must be up-to-date with tetanus and hepatitis vaccinations.
  • Field Safety: When sampling near busy Manila thoroughfares, a spotter must be assigned to monitor traffic.
  • Weather: Sampling should be suspended during heavy typhoon rains or flash flood warnings common in the region.

The Environmental Engineer must prepare the following equipment prior to deployment:

Item Specification Quantity
Sampling Bottles 1L HDPE (for metals), 250mL Glass (for organics) 20 sets
Portable Multiparameter Meter Calibrated for pH, DO, Conductivity, Turbidity 1 unit
Cooler Box Insulated with ice packs (maintain 4°C) 2 units
GPS Device For precise location logging in Manila 1 unit
Preservatives Sulfuric acid (H2SO4), Sodium Thiosulfate As needed

5.1 Site Selection

The Environmental Engineer shall select sampling stations based on the Department of Environment and Natural Resources (DENR) guidelines. Priority areas in Philippines Manila include:

  • Upstream areas near Quezon City.
  • Midstream industrial zones near Mandaluyong and Makati.
  • Downstream areas near Manila Bay.

5.2 Field Sampling Procedure

  1. Calibration: Calibrate the multiparameter meter using standard buffers before leaving the laboratory.
  2. Approach: Approach the sampling site upstream to avoid disturbing the sediment.
  3. In-situ Measurements: Record pH, Dissolved Oxygen (DO), Temperature, and Turbidity immediately upon arrival. Log these values in the field notebook.
  4. Collection: Rinse sampling bottles three times with the site water. Collect samples at a depth of 30cm below the surface to avoid surface film and floating debris.
  5. Preservation:
    • Add Sodium Thiosulfate to samples for BOD analysis to neutralize chlorine.
    • Add Sulfuric Acid to samples for heavy metal analysis to lower pH below 2.
  6. Labeling: Label each bottle with the Station ID, Date, Time, and Sampler's Name.

5.3 Transportation

Immediately place all samples in the cooler box with ice packs. The Environmental Engineer must ensure samples are transported to the accredited laboratory in Manila within 6 hours for biological parameters and 24 hours for chemical parameters to prevent degradation.

Upon arrival at the laboratory, the Environmental Engineer will oversee the analysis of the following parameters using standard methods (APHA/AWWA/WEF):

  • pH: Electrode method.
  • BOD5: 5-day incubation at 20°C.
  • Fecal Coliform: Membrane filtration technique.
  • Heavy Metals: Atomic Absorption Spectroscopy (AAS) for Lead, Mercury, and Cadmium.
  • Turbidity: Nephelometric method.

To ensure data integrity, the following QA/QC measures are mandatory:

  • Field Blanks: One field blank must be collected per sampling trip to check for contamination during transport.
  • Duplicates: 10% of samples must be collected as duplicates to assess precision.
  • Chain of Custody: A signed Chain of Custody form must accompany all samples from the field to the laboratory.

The Environmental Engineer will compile the results into a comprehensive report. The report must compare findings against the DENR Administrative Order No. 2016-08 (Water Quality Guidelines and Standards). Any exceedance of standards must be highlighted, and recommendations for remediation in the Philippines Manila area must be provided.

All chemical waste generated during this experiment must be disposed of according to the Philippines Ecological Solid Waste Management Act. Do not pour chemical residues back into the river or municipal drains.

End of Protocol
